Gambit's a very strong character. "Strong" as far as the depth, detail, history, and complexity of the character. Though essentially an ensemble performance, I see the first two X-Men movies as focusing more on the Wolverine character (another very "strong" character) than the others. Would there be room enough in one movie (time and screen-wise) for two such strong characters?
I've noticed that they've done a fairly good job with foreshadowing in the two movies so far. Characters initially introduced in the first movie were further developed and given more screen time in the second. The Dark Phoenix storyline that seems to be the focus of X3 was hinted at in X2. To see Gambit in X4, would we have to see him first appear in X3?
According to the listed credits, there's no Gambit at all in X3

I hope they don't have a 4th movie. I think they should just do three and then leave it alone. Maybe a spinoff Wolverine movie would be good but I think it's going to be harder to get Patrick Stewart and Ian McCellan to keep comming back. My favorite theme in the X-men story line is tolerance. So even if they put Gambit in, whose mutant power is to be fruity and annoying beyond measure, I will tolerate him.

Perhaps they'll combine McCoy's drinking the serum as part of helping to defeat the Dark Phoenix instead of them being the two separate incidences that they were in the comic series.

There was an article on X3 in my EW that I got today. I can't believe that this is the last movie that they have planned, spin-off's aside! I was hoping that they'd fully flesh out the Phoenix/Bishop/Age-of-Apocalypse storyline... or at least touch upon it more than one film would allow.
Although I shouldn't be surprised, what with the trouble they had even being able to make this one what with getting Halle Berry to resume her role of Storm and other things of that like. Still... the final realization of this is very disappointing.
